α-D-Galactosidase is an exoglycosidase capable of catalyzing the hydrolysis of α-D-galactosidic bonds. In the field of microbiology, this enzyme is widely present in the genus Enterococcus, Salmonella, Klebsiella, and some Escherichia coli. Due to its highly specific expression in certain bacterial genera, α-D-galactosidase activity detection has become one of the "gold standards" for modern microbial chromogenic identification. Through specific chromogenic substrates, direct visual interpretation of target colonies can be achieved, eliminating the need for cumbersome biochemical confirmation steps.
α-D-Galactosidase chromogenic substrates are composed of an indolyl group connected to α-D-galactose via a glycosidic bond. Under the action of α-D-galactosidase produced by bacteria, the glycosidic bond in the substrate molecule is specifically hydrolyzed, releasing the indolyl group. The released indolyl group undergoes oxidative dimerization in air, rapidly forming a visible, water-insoluble colored precipitate.
