β‑D‑Galactosidase (β‑D‑Galactosidase, abbreviated as β‑Gal / LacZ) specifically hydrolyzes β‑D‑galactosidic bonds, making it one of the most classic marker enzymes in molecular biology and microbiology. Based on specialized substrates for β‑Gal , researchers can convert enzyme activity into visible color or fluorescent signals: enabling colony coloration and rapid interpretation on plates/medium, or performing more sensitive signal reading and quantitative analysis in microplate systems.

· Before the reaction:
The substrate molecule is intact, showing no color or fluorescence signal.
· Under the action of β-D-galactosidase:
The β-D-galactosidic bond is specifically cleaved, releasing indole-based or fluorescent signal molecules.
· After the reaction:
For colorimetric substrates, further oxidation or coupling forms a stable colored precipitate;
For fluorescent substrates, high signal-to-noise ratio fluorescence signals are released.
