β‑D‑Galactosidase (β‑Gal / LacZ)


β‑D‑Galactosidase (β‑D‑Galactosidase, abbreviated as  β‑Gal / LacZ) specifically hydrolyzes  β‑D‑galactosidic bonds, making it one of the most classic marker enzymes in molecular biology and microbiology. Based on specialized substrates for β‑Gal , researchers can convert enzyme activity into  visible color or fluorescent signals: enabling colony coloration and rapid interpretation on plates/medium, or performing more sensitive signal reading and quantitative analysis in microplate systems.

1. What is β-D-Galactosidase?

β-D-Galactosidase is a glycoside hydrolase that specifically hydrolyzes β-D-galactosidic bonds and is widely present in various bacteria, with E. coli and Enterobacteriaceae being the most typical examples.
In the field of microbiological detection, β-D-galactosidase activity is often used as a metabolic phenotype indicator to differentiate microbial populations or assist in species identification. β‑Gal substrate-based detection offers three typical advantages:

1. Specificity: Substrate design targets β‑D‑galactosidic bonds, offering more controllable background.

2. Visualizability: Colorimetric substrates can directly present colony color.

3. Quantifiability: Fluorescent/red signal substrates can be used for low-abundance detection, microplate assays, and high-throughput screening.



2. Detection Principle

How β-D-galactosidase substrates generate signals hinges on the introduction of a signal group in their molecular structure that responds to enzymatic cleavage. β-D-galactosidase substrates typically consist of signal group + β-D-galactosidic bond:

· Before the reaction:
The substrate molecule is intact, showing no color or fluorescence signal.

· Under the action of β-D-galactosidase:
The β-D-galactosidic bond is specifically cleaved, releasing indole-based or fluorescent signal molecules.

· After the reaction:
For colorimetric substrates, further oxidation or coupling forms a stable colored precipitate;
For fluorescent substrates, high signal-to-noise ratio fluorescence signals are released.

This reaction system has strong specificity and low background interference, making it suitable for the construction of chromogenic media and the development of enzymatic activity detection methodologies.
